What initially drew you to studying neurodegenerative diseases?<\/span><\/i><\/b><span data-ccp-props=\"{&quot;335559738&quot;:240,&quot;335559739&quot;:120}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">During my undergraduate degree in biochemistry, I became particularly interested in how subtle biochemical changes can\u00a0ultimately lead\u00a0to large-scale effects, such as\u00a0the devastating disease caused by\u00a0single\u00a0amino acid mutations.\u00a0I did my undergraduate project in the lab of Professor Stephen Brickley, and I began learning about neuronal computations that occur in the brain during vision.\u00a0My journey into neuroscience really began with a fascination for how the brain works and what happens when such a complex system goes wrong. What drew me specifically to neurodegeneration was the combination of scientific complexity and human impact. Disorders like Parkinson\u2019s disease and Multiple System Atrophy are devastating for patients and families, and\u00a0despite the advances we have made, we still lack\u00a0meaningful\u00a0disease-modifying treatments.<\/span><span data-ccp-props=\"{&quot;134233117&quot;:true,&quot;134233118&quot;:true,&quot;201341983&quot;:0,&quot;335559740&quot;:240}\">\u00a0<\/span><\/p>\n<p><b><i><span data-contrast=\"none\">Your PhD research focuses on the strain hypothesis of alpha-synuclein in Multiple System Atrophy and Parkinson&#8217;s disease. Could you explain what the strain hypothesis is and why understanding it is so crucial for these conditions?<\/span><\/i><\/b><span data-ccp-props=\"{&quot;335559738&quot;:240,&quot;335559739&quot;:120}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">The strain hypothesis proposes that misfolded alpha-synuclein, the protein that forms the aggregates found in Parkinson\u2019s disease and Multiple System Atrophy, can adopt distinct structural\u00a0configurations, or \u201cstrains.\u201d These strains may differ in how they spread, which cell types they affect, and\u00a0the amount of damage they can do.\u00a0This is particularly important because although\u00a0in\u00a0both conditions\u00a0we see aggregates of the same protein,\u00a0these diseases\u00a0are\u00a0very\u00a0different\u00a0clinically and pathologically. In Parkinson\u2019s disease, alpha-synuclein primarily accumulates in neurons,\u00a0whereas\u00a0in\u00a0Multiple System Atrophy\u00a0it\u00a0predominantly accumulates\u00a0in\u00a0another\u00a0type of\u00a0cell,\u00a0oligodendrocytes. The strain hypothesis offers\u00a0a possible explanation\u00a0for this divergence.\u00a0<\/span><b><span data-contrast=\"auto\">Understanding whether different conformations drive different diseases could transform diagnostics and therapeutic strategies. If we can define disease-specific strains, we may be able to design more targeted interventions rather than treating\u00a0these diseases\u00a0as a single entity.<\/span><\/b><span data-ccp-props=\"{&quot;134233117&quot;:true,&quot;134233118&quot;:true,&quot;201341983&quot;:0,&quot;335559740&quot;:240}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"none\">What makes this area of research particularly exciting or challenging?<\/span><span data-ccp-props=\"{&quot;335559738&quot;:240,&quot;335559739&quot;:120}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">What makes it exciting is that\u00a0we\u2019re\u00a0operating\u00a0at the frontier of structural biology, neuropathology, and disease modelling. New imaging and biochemical techniques are allowing us to interrogate protein aggregation in ways that\u00a0weren\u2019t\u00a0possible even a decade ago. The challenge is that\u00a0we\u2019re\u00a0studying incredibly complex biological systems using post-mortem human tissue, which carries variability and limitations. Interpreting heterogeneity, distinguishing meaningful biological differences from noise, requires both technical rigour and conceptual flexibility. But that complexity is also what makes the discoveries so rewarding.<\/span><span data-ccp-props=\"{&quot;134233117&quot;:true,&quot;134233118&quot;:true,&quot;201341983&quot;:0,&quot;335559740&quot;:240}\">\u00a0<\/span><\/p>\n<p><b><i><span data-contrast=\"none\">Are there any\u00a0technical\u00a0challenges or breakthroughs\u00a0you\u2019ve\u00a0encountered\u00a0in your work?<\/span><\/i><\/b><span data-ccp-props=\"{&quot;335559738&quot;:240,&quot;335559739&quot;:120}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Working with human post-mortem brain tissue presents unique technical challenges, such as\u00a0preservation quality, regional variability, and the need for\u00a0highly sensitive\u00a0detection methods. Optimising immunohistochemical and biochemical protocols to reliably detect different conformations of alpha-synuclein has been a significant part of my work.\u00a0One particularly rewarding breakthrough has been refining approaches that allow us to\u00a0visualise\u00a0the\u00a0functional ability of\u00a0individual\u00a0aggregates within tissue to\u00a0\u201cseed\u201d by\u00a0recruiting\u00a0more protein and growing.\u00a0<\/span><span data-ccp-props=\"{&quot;134233117&quot;:true,&quot;134233118&quot;:true,&quot;201341983&quot;:0,&quot;335559740&quot;:240}\">\u00a0<\/span><\/p>\n<p><b><i><span data-contrast=\"none\">Working in both Professor Steve Gentleman&#8217;s and Professor Javier Alegre-Abarrategui\u2019s\u00a0laboratories must provide diverse perspectives. How do you navigate and benefit from these research environments?\u00a0<\/span><\/i><\/b><span data-ccp-props=\"{&quot;335559738&quot;:240,&quot;335559739&quot;:120}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Working across both Professor Steve Gentleman\u2019s and Professor Javier Alegre-Abarrategui\u2019s\u00a0laboratories has been incredibly enriching. The Gentleman lab brings deep\u00a0expertise\u00a0in neuropathology and human tissue analysis, while the Alegre-Abarrategui\u00a0lab offers strong mechanistic\u00a0perspectives of protein aggregation,\u00a0which aid the development of novel tools to detect early aggregation events.\u00a0Navigating both environments has taught me adaptability,\u00a0understanding how to communicate and integrate different methodological approaches.\u00a0It\u2019s\u00a0strengthened my ability to think about disease from multiple angles,\u00a0from tissue-level pathology to molecular mechanisms.\u00a0This experience\u00a0has shaped the way I design experiments and interpret findings.<\/span><span data-ccp-props=\"{&quot;134233117&quot;:true,&quot;134233118&quot;:true,&quot;201341983&quot;:0,&quot;335559740&quot;:240}\">\u00a0<\/span><\/p>\n<p><b><i><span data-contrast=\"none\">What advice would you give to\u00a0other young\u00a0women who are interested in pursuing careers in medical research?\u00a0<\/span><\/i><\/b><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">I would say,\u00a0don\u2019t\u00a0wait until you feel \u201cready\u201d.\u00a0<\/span><b><span data-contrast=\"auto\">Research is something you grow into<\/span><\/b><span data-contrast=\"auto\">. Confidence often follows experience, not the other way around.\u00a0Seek mentors and collaborators who value your ideas and support your development. Science thrives on diversity of thought, and your perspective is an asset.\u00a0Also,\u00a0don\u2019t\u00a0ever\u00a0stop asking questions;\u00a0curiosity is a powerful driver in research.<\/span><span data-ccp-props=\"{&quot;134233117&quot;:true,&quot;134233118&quot;:true,&quot;201341983&quot;:0,&quot;335559740&quot;:240}\">\u00a0<\/span><\/p>\n<p><b><i><span data-contrast=\"none\">What do you do outside of your professional life to decompress and\u00a0maintain\u00a0balance?\u00a0<\/span><\/i><\/b><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">I try to\u00a0maintain\u00a0balance by stepping completely outside the lab environment when I can,\u00a0whether\u00a0that\u2019s\u00a0spending time\u00a0outdoors or\u00a0catching up with friends and family. Having activities that are unrelated to research helps reset my perspective and\u00a0maintain\u00a0creativity.\u00a0I\u2019ve\u00a0found that some of my best ideas\u00a0emerge\u00a0when\u00a0I\u2019m\u00a0not actively thinking about experiments.<\/span><span data-ccp-props=\"{&quot;134233117&quot;:true,&quot;134233118&quot;:true,&quot;201341983&quot;:0,&quot;335559740&quot;:240}\">\u00a0<\/span><\/p>\n<p><b><i><span data-contrast=\"none\">Is there any forthcoming research project\u00a0you\u2019d\u00a0like to\u00a0mention?<\/span><\/i><\/b><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">I\u2019m\u00a0currently working on further characterising alpha-synuclein aggregate heterogeneity in Multiple System Atrophy using complementary histological and biochemical approaches.
